KrosFlo® Perfusion Systems

Designed for Lab, Pilot and Production Scale Perfusion Operations

KML 100 System for 1.5-100L Bioreactor Volumes
Main Image

KML [KrosFlo MagLev] Benchtop Perfusion and Tangential Flow Filtration (TFF) System is designed for both upstream and downstream TFF process operations. With upstream bioreactor perfusion, the KML is designed for managing 1.5 L to 100L working volume bioreactors using gamma sterilized single-use perfusion flow-paths (MBT's). The system uses a single-use, non-pulsating, levitating magnetic pump controlled by the PLC. The pump is low shear for improved cell viability. The KML system has a small benchtop footprint and comes with a detachable flow path stand.

The KML is a completely integrated system with a color touchscreen to display pressures, flows, weights and process overview with end user set-point functionality. There are visual and audible alarms with safety interlocks to ensure process parameters are maintained. Operators are able to input values for managing six (6) different TFF modes and a manual operation mode. The system comes with two (2) auxiliary peristaltic pumps and two (2) auxiliary scales for precise the management of the flow dynamics are sold separately. Scale Sizes are selected by the user. The non-invasive components include a retentate control valve that manages TMP by adjusting the pinch distance between the plunger and the tubing, and an ultrasonic retentate flow meter that clamps on the tubing. To prevent cells from settling and forming a gel layer on the membrane lumen, the KML system adjusts through a PID control loop from the flow meter.

Further advancements include automatic back pulsing features, USB port for real time data collection, as well as graphical display of process data. KML modes of operation include Perfusion Mode and 5 downstream traditional TFF Modes:

  1. Concentration
  2. Concentration/Diafiltration
  3. Concentration/Diafiltration/Final Concentration
  4. Concentration/Diafiltration(1)/Diafiltration (2)/Final Concentration
  5. Continuous Feed or Diafiltration

KPS® 200 System for 1.5 -100L Bioreactor Volumes

Spectrum developed two KrosFlo Bioreactor Perfusion Systems: the KPS 200 and KPS 600 for upstream bioprocessing. The KPS 200 system can be used with 1.5 -100L liter bioreactors, and the KPS 600 can be used with 100 - 2000 liter bioreactors. The KPS Systems—using Spectrum's sterilized perfusion flow-path assemblies—will automatically control and maintain critical operational parameters defined by the operator (Recirculation Shear Rate, TMP control, Permeate Flow/VVD and Reactor Volume Maintenance). Controls are performed without mechanical components contacting product. Traditional stainless systems need to be CIP or SIP prior to use, while the KPS 200 and KPS 600 are simply loaded with the sterile flow-path assemblies required for the tangential flow perfusion operation. Robust integrated solutions and associated connections minimizes the hold up volume and provides a completely drainable process loop for individual filters ranging with surface areas from 0.047 m2 to 1.0 m2 for the KPS 200 and 0.78 m2 to 12.5 m2 for the KPS 600.

  • KrosFlo ® Perfusion System Components
  • System components include:
  • Software: Proficy HMI/SCADA-IFIX
  • Ethernet port for connection to central data acquisition system
  • 15" Color Touch Screen Computer/Display
  • Sequences Available in Semi-Automated or Automated
